目的分析胃癌发生发展过程中DNA和染色体的不稳定性。方法用 9个随机引物对胃癌DNA做随机引物扩增多态性DNA(RAPD)分析。结果 5号和 3号胃癌样本显示出最高的遗传变异。每个随机引物的检出率有显著差异 ,分别从 2 1%到 85 %不等。结论遗传不稳定性往往集中发生在胃癌细胞染色体上的一些特定位点如微卫星DNA的重复序列上。由于在胃癌的发生发展中有大量位点的遗传物质损伤、变异 ,提示针对单个癌基因或抑癌基因的基因疗法是不易取得成功的。
Objective To analyze the DNA and chromosomal instability in the development of gastric cancer. Methods Random amplified polymorphic DNA (RAPD) analysis was performed on DNA of gastric cancer using nine random primers. Results 5 and 3 gastric cancer samples showed the highest genetic variation. The detection rate of each random primer was significantly different, ranging from 21% to 85%. Conclusion Genetic instability often focuses on some specific sites on the chromosome of gastric cancer cells, such as the repetitive sequences of microsatellite DNA. Due to a large number of genetic material damage and mutation in the development and progression of gastric cancer, gene therapy targeting single oncogene or tumor suppressor gene is not easy to be succeeded.
