藜芦醛和海因经缩合、氢化还原、水解和酯化得到消旋的3,4-二甲氧基苯丙氨酸乙酯,继而在5-硝基水杨醛催化下,经D-(+)-二苯甲酰酒石酸动态动力学拆分得到(S)-(-)-3,4-二甲氧基苯丙氨酸乙酯D-(+)-二苯甲酰酒石酸盐,最后经水解、脱保护得到左旋多巴,条件温和,操作简单,总收率约46.5%。
After condensation, hydrogenation, hydrolysis and esterification of veratraldehyde and hydantoin to racemic 3,4-dimethoxy-phenylalanine ethyl ester, followed by 5-nitro-salicylaldehyde catalyzed by D- (-) - 3,4 - dimethoxy - phenylalanine ethyl ester D - (+) - dibenzoyl tartrate was obtained by dynamic kinetic resolution of (+) - dibenzoyltartaric acid. Finally, hydrolyzed, deprotected to give levodopa, mild conditions, simple operation, the total yield of about 46.5%.
