目的:从体内和体外实验探究茅莓及其总皂苷的抗白血病肿瘤效应。方法:应用已建立的荷白血病瘤裸鼠模型,观察不同浓度茅莓水煎液治疗荷白血病瘤裸鼠后瘤体体积、重量、组织病理的影响;半固体琼脂培养观察不同浓度的含药血清和茅莓总皂苷对K562细胞株集落生长的影响;MTT法观察不同浓度的含药血清和茅莓总皂苷对K562细胞株的增殖作用的影响。结果:低中高剂量茅莓水煎液(0.5g/mL,1g/mL,2g/mL)治疗荷瘤裸鼠后,瘤重分别为(0.95±0.13)g、(0.49±0.09)g和(0.33±0.01)g,抑瘤率分别为57.55%、75.40%和87.16%,抑制效应呈剂量依赖性方式;HE染色治疗组瘤体内可见多个坏死灶,瘤细胞肿胀、破裂,细胞核溶解、消失;琼脂半固定集落培养发现不同浓度含药血清及茅莓总皂苷均能抑制K562细胞株集落形成,抑制效应呈剂量依赖性方式,其中终浓度为10%和20%的含药血清抑制集落形成率为(21.6±2.2)%和(48.8±1.2)%,与对照组相比差异有统计学意义(P均<0.05),20 mg/L茅莓总皂苷抑制集落形成率为(22.5±1.9)%,与对照组相比差异有统计学意义(P<0.05),且当浓度增加至150mg/L时,无集落形成;MTT示不同浓度的含药血清及茅莓总皂苷均能抑制K562细胞株的增殖,呈剂量依赖性方式,其中20%的含药血清可明显抑制K562细胞增殖,与对照组相比差异有统计学意义(P<0.05),20mg/L浓度的茅莓总皂苷能明显的抑制K562增殖,与对照组相比差异有统计学意义(P<0.05)。结论:茅莓水煎剂对荷白血病瘤裸鼠瘤体生长有明显抑制作用,且一定浓度的含药血清和茅莓总皂苷对K562集落形成和增殖均有明显的抑制作用,为临床用药提供了一定的理论依据。
OBJECTIVE: To explore the anti-leukemia tumor effect of raspberry and its total saponins from in vitro and in vivo experiments.
