目的 研究MxA抗病毒蛋白的表达及其活性。方法 用IFNα2b处理WI - 38细胞或人外周血单核细胞 12h或 2 4h ,并用Western -blot法或FACS法分别进行MxA蛋白表达的检测及微量细胞病变抑制法对重组的MxA和IFNα2b进行抗病毒效应实验。结果 (1)低浓度IFNα2b(>1IU/ml)可诱导WI- 38细胞表达MxA ;(2 ) 10ng/mlMxA可以抵抗 2 0TCID50 的HSV -I、Polio .V感染Vero细胞 ,抵抗 2 0 0TCID50 的VSV感染Wish细胞。结论 MxA是由I型干扰素特异诱导产生的 ,且具有广谱的直接抗病毒活性。
Objective To study the expression of MxA antiviral protein and its activity. Methods WI - 38 cells or human peripheral blood mononuclear cells were treated with IFNα2b for 12 hours or 24 hours. The expression of MxA protein was detected by Western - blotting or FACS, and the anti - virus activity of recombinant MxA and IFNα2b Effect experiment. Results (1) Low concentration of IFNα2b (> 1IU / ml) induced MxA expression in WI-38 cells. (2) 10ng / ml MxA could resist 20TCID50 of HSV-I and PolioV infected Vero cells and resistant to 200TCID50 of VSV Infection Wish cells. Conclusion MxA is specifically induced by type I interferons and has a broad spectrum of direct antiviral activity.
