目的 建立一种成骨细胞体外培养模型。方法 以新生兔颅骨为材料 ,用改良组织块移行生长法在体外培养获得贴壁生长的细胞 ,并对其形态 ,生长特点及生物学特性进行研究。结果 获得的细胞呈多种生长形态 ,如梭形 ;三角形 ;鳞片形 ;星形等 ,有较多的细胞突起 ,细胞呈复层生长 ,互相重叠 ,形成细胞结节 ,碱性磷酸酶(AL P)染色强阳性 ,在含有 Vit C和 β—甘油磷酸钠的培养条件中出现钙化现象。结论 采用改良组织块移行生长法获得的细胞具有成骨细胞的特性 ,该方法是体外培养、研究成骨细胞的良好模型。
Objective To establish an osteoblast culture model in vitro. Methods Newborn rabbit skull was used as material, and adherent cells were cultured in vitro by modified tissue block migration method. The morphology, growth characteristics and biological characteristics of the cells were studied. Results The cells obtained in various growth patterns, such as fusiform; triangle; scale-shaped; star, etc., more cells protruding, the cells were stratified growth, overlapping, forming cell nodules, alkaline phosphatase P) strongly positive, calcification occurred in culture conditions containing Vit C and β-glycerophosphate. Conclusions The cells obtained by the method of modified tissue block migration growth have the characteristics of osteoblasts. This method is a good model for osteoblast culture in vitro.
