目的研究麻疯树Jatropha curcas种子中具有抗氧化活性的有效成分。方法使用DPPH活性测试方法进行抗氧化活性的筛选和追踪,采用活性追踪性分离方法,使用硅胶、ODS和凝胶柱色谱等现代分离手段分离得到麻疯树种子中具有抗氧化活性的有效成分。综合应用多种光谱学方法(UV、IR、ESI-MS、HR-ESI-MS、1H-NMR、13C-NMR、2D-NMR)对所得化合物进行解析并鉴定其结构。最后使用DPPH法测定并评价单体化合物的抗氧化活性。结果麻疯树种子提取物的正丁醇萃取层具有显著的抗氧化活性,通过对该萃取部分的成分研究共分离得到6个化合物,经结构解析,分离得到的6个化合物分别鉴定为麻疯果素A(3,4,4′5′-四羟基-3′-甲氧基-双环氧木脂素,3,4,4′,5′-tetrahydroxyl-3′-methoxylbisepoxylignan,1)、(±)-3,3′-bisdemethylpinoresinol(2)、7-表-芝麻素-二儿茶酚(3)、异巴西油大戟素(isoprincepin,4)、β-谷甾醇(5)与β-胡萝卜苷(6)。结论活性测定结果表明,化合物3、4具有较强活性,化合物1、2具有中等强度活性。化合物1为新化合物,命名为麻疯果素A(jatropha-sin A);化合物2、3、4为首次从麻疯树属中分离得到。
Objective To study the antioxidant active ingredients in Jatropha curcas seeds of Jatropha curcas. Methods Antioxidant activity was screened and traced using DPPH activity test method. The active components of Jatropha curcas seeds were separated by active tracing separation method and separated by modern separation methods such as silica gel, ODS and gel column chromatography. The compounds were resolved and their structures identified using a variety of spectroscopic methods (UV, IR, ESI-MS, HR-ESI-MS, 1H-NMR, 13C-NMR, 2D-NMR) Finally, the DPPH method was used to determine and evaluate the antioxidant activity of monomeric compounds. Results The extract of Jatropha curcas seeds showed n-butanol-extractable layer. Six compounds were isolated from the constituents of the extracted fraction. The six compounds isolated from the extracts were identified as leprosy Fructose A (3,4,4’5’-tetrahydroxy-3’-methoxy-digoxyignan, 3,4,4 ’, 5’-tetrahydroxyl-3’-methoxylbisepoxylignan, 1) (3), isoprincepin (4), β-sitosterol (5) and β-sitosterol - daucosterol (6). Conclusion The results of activity assay showed that compounds 3 and 4 have strong activity and compounds 1 and 2 have moderate activity. Compound 1 is a new compound, named jatropha-sin A; Compounds 2, 3 and 4 were isolated from Jatropha for the first time.
