用MTT法检测了硫酸软骨素A(4-硫酸软骨素,CS-A)对NIH-3T3鼠成纤维细胞和人皮肤成纤维细胞存活与增殖的影响。发现所侧OD值随CS-A浓度的增高而增大,两者呈正相关(3T3鼠成纤维细胞:r=0.8154,P<0.05;人皮肤成纤维细胞:r=0.915,P<0.01),表明CS-A对这两类成纤维细胞的存活和增殖有促进作用。采用PAP和ABC免疫细胞化学染色,观察到两类细胞的胞膜上有CS分布,提示CS可能参与膜结构的组成,对细胞-细胞与细胞-基质之间的相互作用有重要意义;为进一步探讨硫酸软骨素在肝纤维化中的作用提供了可靠的实验依据。
The effects of chondroitin sulfate A (chondroitin sulfate-4, CS-A) on the survival and proliferation of NIH-3T3 murine fibroblasts and human dermal fibroblasts were examined by MTT assay. It was found that the OD value increased with the increase of CS-A concentration, the two were positively correlated (3T3 murine fibroblasts: r = 0.8154, P <0.05; human skin fibroblasts: r = 0.915 , P <0.01), indicating that CS-A can promote the survival and proliferation of these two types of fibroblasts. Using PAP and ABC immunocytochemical staining, it was observed that the distribution of CS on the cell membrane of both cell types suggests that CS may participate in the membrane structure and is of great importance to the cell-cell-cell-matrix interaction. To further To explore the role of chondroitin sulfate in liver fibrosis provides a reliable experimental basis.
