为研究检测人肿瘤组织细胞多药耐药基因表达产物,采用间接标记法,用鼠抗人P170单克隆抗体,二抗为FITC标记兔抗鼠Ig,应用流式细胞仪检测了29例新鲜实体肿瘤组织和一株多药耐药的K562细胞株。结果显示:18例肿瘤细胞表达P170,其阳性率为62.1%,11例未检出表达,其阴性率为37.9%;99.9%K562细胞表达P170。在18例P170阳性表达的样本中,有16例阳性表达百分比低于30%;只有2例高于30%。提示大多数肿瘤组织细胞表达P170阳性率高,但表达P170肿瘤细胞表达百分比低。检测肿瘤组织的P170表达能为临床治疗提供一个可靠的参考指标。
In order to study the detection of multidrug resistance gene expression products in human tumor cells, indirect labeling was used. Mouse anti-human P170 monoclonal antibody and secondary antibody FITC were used to label rabbit anti-mouse Ig. 29 fresh entities were detected by flow cytometry. Tumor tissue and a multidrug resistant K562 cell line. The results showed that: 18 cases of tumor cells expressed P170, the positive rate was 62.1%, 11 cases were not detected, the negative rate was 37.9%; 99.9% K562 cells express P170. Of the 18 samples with positive P170 expression, 16 were positive for less than 30%; only 2 were higher than 30%. It is suggested that the majority of tumor cells express P170 with high positive rate, but express P170 tumor cells with low expression percentage. Detecting P170 expression in tumor tissue can provide a reliable reference for clinical treatment.
