采用鸡枞菌多糖(TAP)体外作用于小鼠T淋巴细胞,用ELISA法检测其IL-4、IFN-γ及IL-2水平,流式细胞仪检测T淋巴细胞亚群,RT-PCR法检测相关细胞因子mRNA表达量,观察研究了鸡枞菌多糖对小鼠T细胞免疫功能的影响。结果表明,与ConA组相比,25~100μg/mL TAP在作用72h内均能显著促进T细胞产生IL-4、IFN-γ及IL-2(P<0.01);25μg/mL TAP作用48h后可显著降低被ConA激活的T细胞CD4+%(P<0.01),升高CD8+%(P<0.05),并降低CD4+/CD8+(P<0.05);50μg/mL TAP则可显著升高CD3+%(P<0.01);50μg/mL TAP在作用72h内能显著提高IL-2、IL-4及IFN-γmRNA表达量(P<0.01),并使其保持较高水平。提示,TAP对细胞免疫功能具有显著增强作用,T淋巴细胞是TAP的靶细胞之一。
T-lymphocytes were treated with Termitomyces polysaccharides (TAP) in vitro. The levels of IL-4, IFN-γ and IL-2 were detected by ELISA. T lymphocyte subsets were detected by flow cytometry. The expression of related cytokines was detected and the effect of Termitomyces polysaccharide on T cell immune function was observed. The results showed that compared with the ConA group, 25 ~ 100μg / mL TAP could significantly promote the production of IL-4, IFN-γand IL-2 in T cells within 72h (P <0.01) (P <0.01), increase CD8 +% (P <0.05), and decrease CD4 + / CD8 + (P <0.05). Conversely, 50μg / mL TAP can significantly increase CD3 +% (P < P <0.01). The expression of IL-2, IL-4 and IFN-γmRNA of TAP at 50μg / mL within 72h significantly increased (P <0.01) and kept at a higher level. Tip, TAP has a significant enhancement of cellular immune function, T lymphocytes is one of TAP target cells.
