目的探讨水通道蛋白AQP1-AQP9,AQP11及AQP12在多种肿瘤细胞系中的表达并探讨其意义。方法利用RT-PCR方法对小鼠乳腺癌细胞系4T1、mmt和lewis肺癌细胞系(LLC)表达水通道情况进行初步分析。结果4T1细胞表达AQP5、AQP7及AQP11,AQP5只有少量表达,后两者表达水平较高;在lewis肺癌(LLC)中有AQP7,AQP8及AQP11表达,其中AQP7和AQP11表达较高;在mmt细胞中AQP3、AQP4、AQP7及AQP11均被检出有表达,AQP7表达程度较高。结论肿瘤细胞表达水通道蛋白是一普遍现象,可能与肿瘤细胞易转移和高度增殖等特性有关。深入研究水通道蛋白在肿瘤细胞增殖及迁移中的作用,将为肿瘤的发生、发展及转移提供新的机理。
Objective To investigate the expression of aquaporin AQP1-AQP9, AQP11 and AQP12 in various tumor cell lines and to explore its significance. Methods The expression of water channels in mouse breast cancer cell lines 4T1, mmt and lewis lung cancer cell lines (LLC) were analyzed by RT-PCR. Results AQP5, AQP7 and AQP11 were expressed in 4T1 cells and only a small amount of AQP5 was expressed in 4T1 cells. The expression levels of AQP7, AQP8 and AQP11 were higher in lewis lung cancer (LLC) AQP3, AQP4, AQP7 and AQP11 were detected expression, AQP7 expression level higher. Conclusion The expression of aquaporin in tumor cells is a common phenomenon, which may be related to the characteristics of tumor cells such as metastasis and hyperplasia. In-depth study of the role of aquaporin in tumor cell proliferation and migration will provide a new mechanism for tumorigenesis, development and metastasis.
