目的 用B2 3蛋白的单克隆抗体研究B2 3表达在鼠肝大部分切除后再生过程中肝细胞的动态变化 ,探讨B2 3作为肝再生和肝细胞增生标记物的可能性。方法 鼠肝大部切除术后第 1、2、3、4和 7d收取再生肝组织 ,采用B2 3的单克隆抗体和Western印迹及免疫组化方法检测B2 3在不同时间再生肝组织的表达 ,并与增殖细胞核抗原 (PCNA)的表达相比较。结果 B2 3在静止期肝细胞中几乎检测不到 ,但在不同时间再生肝组织的表达中既有表达量的变化又有表达位置的变化 ,在术后 1~ 7d再生肝组织的表达中 ,B2 3表达量的增高变化呈近似抛物线样变化 ,其中第 3d表达水平最高 ,B2 3表达量的这些变化与PCNA的变化非常相似。B2 3表达位置的变化从核仁的增强表达 (G1 S)开始 ,进而核浆的表达 (S G2 )及胞质和分裂染色体的表达 (M)。结论 B2 3蛋白表达可作为肝再生和肝细胞增生的标记
Objective To study the dynamic changes of B2 3 expression in hepatocytes during the excision of most of the excised rat hepatocytes with the monoclonal antibody B2 3 and to explore the possibility of B2 3 as a marker of liver regeneration and hepatocyte proliferation. Methods Rat liver tissues were collected at 1, 2, 3, 4 and 7 days after subtotal hysterectomy. B2 3 monoclonal antibodies and Western blotting and immunohistochemistry were used to detect the expression of B2 3 in regenerating liver tissues at different time points. And compared with the expression of proliferating cell nuclear antigen (PCNA). RESULTS: B2 3 was almost undetectable in quiescent liver cells, but there was both expression change and expression change in liver tissue at different time points. Among the liver tissue regenerated from 1 to 7 days after operation, The increase of B2 3 expression showed a parabola-like change with the highest expression level at the third day. The changes of B2 3 expression were similar to those of PCNA. Changes in B2 3 expression start from enhanced nucleolar expression (G1 S), and then from cytoplasm expression (S G2) and cytoplasmic and chromosomal expression (M). Conclusion B2 3 protein expression can be used as a marker of liver regeneration and hepatocyte proliferation
