目的建立复方姜黄素脂质立方液晶纳米粒姜黄素和胡椒碱包封率的测定方法。方法采用HPLC法测定,色谱柱为Kromasil C18柱(4.6 mm×250 mm,4.6μm),流动相为乙腈-0.4%磷酸(48∶52),流速为1 mL.min-1,检测波长为343 nm,430 nm(波长转换时间:16.5 min);采用微柱离心法和透析法测定包封率。结果姜黄素在1.8081~18.081 ng范围内线性良好(r=0.9998);胡椒碱在3.3331~33.331 ng范围内线性良好(r=0.9999);微柱离心法和透析法测定姜黄素、胡椒碱的包封率分别为59.52%、56.02%和60.13%、55.86%。结论采用微柱离心法和HPLC法可以快速、准确地测定复方姜黄素脂质立方液晶纳米粒姜黄素和胡椒碱的包封率。
Objective To establish a method for the determination of the entrapment efficiency of curcumin and piperine in compound curcumin lipid cubic liquid crystal nanoparticles. The method was performed on a Kromasil C18 column (4.6 mm × 250 mm, 4.6 μm) with a mobile phase of acetonitrile-0.4% phosphoric acid (48:52) at a flow rate of 1 mL · min-1 and a detection wavelength of 343 nm, 430 nm (wavelength conversion time: 16.5 min). The entrapment efficiency was determined by microcolumn centrifugation and dialysis. Results Curcumin was linear over the range of 1.8081 ~ 18.081 ng (r = 0.9998). Piperine was linear over the range of 3.3331 ~ 33.331 ng (r = 0.9999). The contents of curcumin and piperine were determined by microcolumn centrifugation and dialysis The seal rates were 59.52%, 56.02% and 60.13%, 55.86% respectively. Conclusion The microcapsule centrifugation and HPLC method can be used to determine the encapsulation efficiency of compound curcumin lipid cubic liquid crystal nanoparticles curcumin and piperine rapidly and accurately.
