目的建立猪Ⅲ度烧伤所致全层皮肤缺损并植入人工真皮支架的动物模型,观察人工真皮支架移植后1、2、3周,支架血管化情况以及人工真皮支架上移植刃厚皮片的可能性。方法造成直径约3 cm的Ⅲ度烧伤创面,48 h后清创后植入胶原-磺化羧甲基壳聚糖/硅橡胶双层人工真皮支架,术后悬吊饲养、隔天换药护理。支架植入后1、2、3周观察创面并植刃厚皮片,植皮2周后观察植皮成活及创面修复。结果①造成约3 cm直径烧伤创面的铜锭最佳直径为2.5 cm;②为防止创面收缩必须应用移植环,手术后必须悬吊饲养;③支架植入后植皮时间最好选择在2周以后;④良好的术后处理是实验成功的重要因素。结论本实验模型稳定、可重复性强,为人工皮肤的研究和生物学意义的验证提供了良好的大型动物模型。
OBJECTIVE: To establish an animal model of full-thickness skin defect caused by third degree burn in pigs implanted with artificial dermal scaffolds and to observe the vascularization of the scaffolds after artificial dermal scaffolds were implanted and the thickness of grafts on the artificial dermal scaffolds possibility. Methods A third degree burn wound of about 3 cm in diameter was created. After 48 h, the wounds were implanted with collagen-sulfonated carboxymethyl chitosan / silicone double-layered artificial dermal scaffolds. . At 1, 2 and 3 weeks after stent implantation, the wounds were observed and covered with thick skin graft. Skin graft survival and wound healing were observed 2 weeks after skin grafting. Results ① The diameter of the copper ingot that caused the burn wound about 3 cm in diameter was 2.5 cm. ② The graft ring must be applied to prevent the wound from shrinking. After the operation, the copper ingots must be suspended and reared. ; ④ good postoperative treatment is an important factor in the success of the experiment. Conclusion The experimental model is stable and reproducible. It provides a good large animal model for the study of artificial skin and the verification of biological significance.
