目的:明确芪参健骨颗粒对激素合并内毒素所致的股骨头坏死模型大鼠骨组织的影响,并探讨其作用机制。方法:SD大鼠随机分为空白组、模型组、低、中、高剂量给药组、阳性对照组6组,除正常组外其余各组均用激素加内毒素诱导建立股骨头坏死模型,造模与给药同时进行,低、中、高剂量组分别给予芪参健骨流浸膏15、30、60g·kg-1,阳性对照组给予丹郁骨康丸3g·kg-1,正常组、模型组给予等量生理盐水,给药8周后,腹主静脉取血测血钙血磷含量;取两侧股骨,左侧用于制作组织病理切片,右侧进行骨密度测定,三点弯曲试验。结果:芪参健骨颗粒低、中、高剂量组及阳性对照组骨密度、最大载荷量、单位面积最大载荷量、弹性模量及血钙、血磷含量均显著高于模型组;模型组骨密度、最大载荷量、单位面积载荷量、弹性模量、血钙血磷含量显著低于正常组。结论:芪参健骨颗粒具有防治股骨头坏死的作用,可能通过提高骨密度、骨强度及血钙血磷含量发挥作用。
OBJECTIVE: To determine the effect of Qi-shen Jiangu Granule on the bone tissue of rats with osteonecrosis of the femoral head caused by hormone and endotoxin, and to explore its mechanism. Methods: Sprague-Dawley rats were randomly divided into blank group, model group, low, medium and high dose groups and 6 positive control groups. All the rats except the normal group were induced by hormones and endotoxin to establish the model of femoral head necrosis. The models were given at the same time. The low, medium and high dose groups were given QCHRG 15, 30, 60g · kg-1, respectively. The positive control group was given Danyu Gukang Pill 3g · kg-1, Group, the model group was given the same amount of saline, after 8 weeks of administration, the blood taken from the main venous blood was measured for serum calcium and phosphorus; two sides of the femur were taken, the left for the production of histopathological sections, the right bone mineral density determination, Point bending test. Results: The bone mineral density, the maximum load, the maximum load per unit area, the elastic modulus, the content of serum calcium and phosphorus in the QSGJ low, middle and high dose groups and the positive control group were significantly higher than those in the model group BMD, maximum load, load per unit area, elastic modulus, serum calcium and phosphorus were significantly lower than those in normal group. Conclusion: Qishen Jiangu Granule can prevent and cure femoral head necrosis and may play its role by increasing bone density, bone strength and serum calcium and phosphorus levels.
