目的建立五生饮中3种单酯型乌头类生物碱(苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱)的含量测定方法。方法采用YMC-Pack ODS-AMC18柱(250 mm×4.6 mm,5μm),柱温:35℃,以乙腈-四氢呋喃(25:15)为流动相A,0.1 mol·L~(-1)醋酸铵溶液(每1000 m L加冰醋酸0.5 mL)为流动相B,梯度洗脱,流速:1.0 m L·min~(-1),检测波长:235 nm,进样量:10μL。结果苯甲酰新乌头原碱在146.96~1469.60 ng范围内线性关系良好,r=0.9999,平均回收率为99.05%(RSD为1.87%);苯甲酰乌头原碱在16.78~167.80 ng范围内线性关系良好,r=0.9998,平均回收率为99.42%(RSD为1.94%);苯甲酰次乌头原碱在20.30~203.00ng范围内线性关系良好,r=0.9999,平均回收率为100.15%(RSD为1.53%)。结论本研究所建立的方法操作简单,稳定性好,准确度高,重复性好,为五生饮水煎剂的质量控制提供参考。
Objective To establish a method for the determination of three monosaccharide aconite alkaloids (benzoyl mesaconitine, benzoyl aconitine and benzoyl aconitine) in Wusheng Yin. Methods The column of YMC-Pack ODS-AMC18 (250 mm × 4.6 mm, 5 μm) was used. The mobile phase A was acetonitrile-tetrahydrofuran (25:15) The mobile phase B was gradient elution at a flow rate of 1.0 mL · L -1, the detection wavelength was 235 nm and the injection volume was 10 μL. Results The linear range of benzoyl mesaconitine in the range of 146.96-1469.60 ng was good (r = 0.9999, the average recovery was 99.05% (RSD = 1.87%)); benzoyl aconitine was in the range of 16.78-167.80 ng (R = 0.9998, the average recovery was 99.42% (RSD = 1.94%)); benzoyl aconitine alkaloids in the range of 20.30 ~ 203.00ng good linearity, r = 0.9999, the average recovery was 100.15 % (RSD 1.53%). Conclusion The method established in this study has simple operation, good stability, high accuracy and good reproducibility. It provides a reference for the quality control of Wusheng drinking decoction.
