目的 :选择并建立异种聚合血红蛋白抗原性检测方法 ,为评价异种聚合血红蛋白对人有无抗原性提供参考。方法 :从速发型超敏反应、体液免疫反应、细胞免疫反应和抗原交叉反应检测聚合牛血红蛋白对异种动物的抗原性。结果 :家兔及豚鼠未发生速发型超敏反应。多次免疫后 ,家兔血清中总IgG无明显变化 ,但特异性IgG水平显著升高。B淋巴细胞转化检测表明 ,B淋巴细胞增殖活性增强 ,但与对照组相比无显著差异 (P >0 .0 5 )。聚合牛血红蛋白、人血红蛋白、兔血红蛋白抗原交叉反应结果显示 ,聚合牛血红蛋白与人血红蛋白存在抗原交叉反应 ,提示聚合牛血红蛋白与人血红蛋白在抗原结构上有同源性。结论 :该聚合牛血红蛋白与人血红蛋白在抗原结构上有同源性 ,除用其免疫的实验组动物血清特异性抗体水平显著升高外 ,其他检测指标无显著变化。根据免疫学理论 ,无论某异物刺激机体产生何种免疫应答反应 ,都应视为该异物存在抗原性 ,因此提示该聚合血红蛋白存在抗原性。
OBJECTIVE: To select and establish a method for detecting the antigenicity of heterogeneous aggregated hemoglobin, and to provide a reference for evaluating the antigenicity of human heterogeneous hemoglobin. Methods: The antigenicity of polymerized bovine hemoglobin (HbAc) against heterologous animals was examined by rapid hair-type hypersensitivity, humoral immune response, cellular immune response and antigen cross-reactivity. Results: No immediate hypersensitivity occurred in rabbits and guinea pigs. After repeated immunization, there was no significant change in total IgG in rabbit serum, but the level of specific IgG was significantly increased. B lymphocyte transformation test showed that B lymphocyte proliferation activity increased, but compared with the control group no significant difference (P> 0.05). The results of cross-reaction of aggregated bovine hemoglobin, human hemoglobin and rabbit hemoglobin antigen showed that there was cross-reaction between polymerized bovine hemoglobin and human hemoglobin, suggesting that homologous aggregation of bovine hemoglobin and human hemoglobin in antigen structure. CONCLUSION: The aggregated bovine hemoglobin and human hemoglobin have homology in antigen structure. There was no significant change in the other indexes except the serum-specific antibody raised in the immunized group. According to immunological theory, no matter what kind of foreign body to stimulate the body to produce immune response, should be regarded as antigens exist in the foreign body, suggesting that there is antigenicity of the aggregated hemoglobin.
