目的制备并评价芦丁-羟丙基-β-环糊精包合物。方法本实验采用冷冻干燥法制备芦丁-羟丙基-β-环糊精包合物,通过L9(3)4正交设计优选投料比、包合时间、包合温度,通过考察包合物的溶解度、相溶解度验证包合物的形成,使用X射线衍射法对包合物进行鉴定。结果正交设计筛选出的最佳条件为投料比1∶4,包合时间2 h,包合温度为45℃。以该法制备的芦丁-羟丙基-β-环糊精包合物溶解度得到显著提高,达到9.61 mg.mL-1。结论本实验采用的包合方法及材料可以显著改善芦丁的溶解性能。
Objective To prepare and evaluate rutin-hydroxypropyl-β-cyclodextrin inclusion complex. Methods In this experiment, the inclusion complex of rutin-hydroxypropyl-β-cyclodextrin was prepared by freeze-drying method. The orthogonal design of L9 (3) 4 was adopted to optimize the feed ratio, inclusion time and inclusion temperature. The solubility and phase solubility of the inclusion complex were tested. The inclusion complex was identified by X-ray diffraction. Results The optimal conditions for the orthogonal design were as follows: feed ratio 1: 4, inclusion time 2 h, inclusion temperature 45 ℃. The solubility of rutin-hydroxypropyl-β-cyclodextrin inclusion complex prepared by this method was significantly improved, reaching 9.61 mg.mL-1. Conclusion The inclusion methods and materials used in this experiment can significantly improve the solubility of rutin.
