本文用荧光薄层扫描法测定庆大霉素发酵液中C组分含量。用氯仿:甲醇:氨水=4:5:5下相 为展开剂,在硅胶G-CMC板上分离庆大霉素C1,C2及C1a,喷甘油作增荧光试剂,然后用薄层 扫描仪测定其荧光强度。本法具有快速、简便、灵敏度高(C1,C2及C1a的检出限量分别为 0.3、 0.7及0.2μg),线性范围宽及重现性好等优点。也可用于成品及半成品测定。
In this paper, the content of C component in gentamicin fermentation broth was determined by fluorescence TLC. Gentamicin C1, C2 and C1a were separated on silica gel G-CMC plate using glycerol: methanol: ammonia = 4: 5: 5 as the developing solvent, then glycerol was sprayed as fluorescent reagent and then measured by TLC scanner Its fluorescence intensity. The method has the advantages of fast, simple and high sensitivity (detection limits of C1, C2 and C1a are 0.3, 0.7 and 0.2μg, respectively), wide linear range and good reproducibility. Can also be used for the determination of finished and semi-finished products.
