目的:观察来氟米特对子宫内膜异位症大鼠模型病灶生长行为的影响,为子宫内膜异位症的免疫治疗提供依据。方法:采用自体移植法建立大鼠子宫内膜异位症模型。移植第3周治疗组给予来氟米特35 mg/kg.d,对照组给予生理盐水1 m l/kg.d,连用3周;测量用药前后病灶体积,并采用放免法检测TNF-α水平,免疫组化法检测NF-κB的表达情况。结果:治疗组病灶体积显著低于对照组(P<0.05),组织病理学显示治疗组的异位内膜生长明显受抑制或变性坏死,病灶组织血供不足;治疗组TNF-α及NF-κB表达显著低于对照组(P<0.05)。结论:来氟米特对大鼠子宫内膜异位病灶的生长有明显抑制作用,证实免疫调节治疗子宫内膜异位症的策略具有可行性。
OBJECTIVE: To observe the effect of leflunomide on the growth of endometriosis rat model and provide evidence for the immunotherapy of endometriosis. Methods: The rat endometriosis model was established by autograft. The third week after transplantation, leflunomide 35 mg / kg.d was given to the treatment group, while the control group was given saline 1 ml / kg.d for 3 weeks. The volume of the lesion was measured before and after treatment. The level of TNF- Immunohistochemistry was used to detect the expression of NF-κB. Results: The lesion volume of the treatment group was significantly lower than that of the control group (P <0.05). Histopathology showed that the ectopic endometrial growth of the treated group was significantly inhibited or degenerated and necrotic, and the blood supply of the lesion was insufficient. The TNF- κB expression was significantly lower than the control group (P <0.05). CONCLUSION: Leflunomide can significantly inhibit the growth of endometriotic lesions in rats, confirming the feasibility of immunomodulatory strategies for the treatment of endometriosis.
