目的观察肝脏X受体(LXR)激动剂TO901317对脂多糖LPS致小鼠中脑内炎症反应的调节作用。方法提取小鼠中脑mRNA行RT-PCR及中脑免疫组织化学方法检测LXR激动剂TO901317对LPS致炎小鼠中脑内炎症因子IL-1β,IL-6和TNF-α的影响及小胶质细胞活化的调节作用。结果 TO901317处理组小鼠中脑IL-1β,IL-6和TNF-αmRNA含量明显低于单纯LPS注射组。TO901317预处理抑制LPS引起的中脑小胶质细胞的活化。结论 LXR受体激活能有效抑制中脑小胶质细胞激活及前炎因子的释放。
Objective To investigate the regulatory effect of hepatic X receptor (LXR) agonist TO901317 on the inflammatory response in mice induced by lipopolysaccharide LPS. Methods The midbrain mRNA was extracted and the effects of LXR agonist TO901317 on the intracerebral inflammatory cytokines IL-1β, IL-6 and TNF-α in LPS-induced mice were detected by RT-PCR and midbrain immunohistochemistry. Modulation of the Activation of Stromal Cells. Results The levels of IL-1β, IL-6 and TNF-αmRNA in midbrain of TO901317-treated mice were significantly lower than those of LPS-injected mice. TO901317 pretreatment inhibits the activation of midbrain microglia induced by LPS. Conclusion LXR receptor activation can effectively inhibit the activation of midbrain microglia and the release of pro-inflammatory cytokines.
