目的探讨邻苯二甲酸二丁酯(DBP)母体暴露对胚胎期大鼠睾丸蛋白表达谱的影响。方法孕SD大鼠10只,随机分为2组,妊娠14~18 d,实验组和对照组分别予DBP每日800 mg/kg体重、大豆油5 ml/d灌胃,第19天取出子鼠睾丸,每组各3例样本,提取总蛋白,行二维凝胶电泳分离和质谱分析。结果筛选出23个差异表达蛋白(t≥2.831,P<0.05),经蛋白数据库检索,共鉴定出差异蛋白14个(评分>50,P<0.05),这些蛋白主要参与细胞凋亡、细胞增殖与分化和信号转导等。结论蛋白质组学是研究DBP的生殖系统发育毒性的有效方法。
Objective To investigate the effect of DBP maternal exposure on the expression of testis protein in embryonic rats. Methods Ten pregnant SD rats were randomly divided into two groups, 14 to 18 days of gestation. The experimental group and the control group were given DBP 800 mg / kg body weight daily and soybean oil 5 ml / Rats testis, each group of 3 samples, total protein extraction, two-dimensional gel electrophoresis separation and mass spectrometry. RESULTS: Twenty-three differentially expressed proteins were screened (t≥2.831, P <0.05). A total of 14 differential proteins (score> 50, P <0.05) were identified by protein database searching. These proteins were mainly involved in apoptosis and cell proliferation With differentiation and signal transduction. Conclusion Proteomics is an effective method to study the reproductive developmental toxicity of DBP.
