目的 探讨术前区域动脉灌注化学药物治疗 (化疗 )对结直肠癌细胞生长周期的影响。方法 采用Seldinger技术 ,经股动脉插管将化疗药注入癌灶供血动脉。化疗结直肠癌 30例 ,术后 10d进行结直肠癌根治术。用免疫组织化学方法测定p16蛋白和Rb蛋白的表达情况 ,与未行化疗的对照组进行比较。结果 术前区域动脉灌注化疗组p16蛋白表达标记指数 (LI)为 (35± 19) % ;对照组为(16± 8) %。化疗组p16蛋白表达明显高于对照组 ,P <0 0 1。术前区域动脉灌注化疗后癌细胞中Rb蛋白表达的LI为 (4 8± 11) % ,明显高于对照组〔LI为 (38± 10 ) %〕 ,P <0 0 1。p16蛋白与Rb蛋白表达与Dukes分期显著相关 ,B期表达较C、D期为高。结论 术前区域动脉灌注化疗可通过上调p16与Rb蛋白表达 ,在G1/S限制点发挥抑制细胞周期进入S期的作用
Objective To investigate the effect of preoperative regional arterial infusion chemotherapy (chemotherapy) on the growth cycle of colorectal cancer cells. Methods Seldinger technique was used to inject chemotherapeutic drugs into the tumor feeding artery through the femoral artery. Thirty patients with chemotherapy and colorectal cancer were treated with radical resection of colorectal cancer 10 days after surgery. The expression of p16 protein and Rb protein was measured by immunohistochemistry and compared with the control group without chemotherapy. Results The preoperative regional arterial infusion chemotherapy group p16 protein expression index (LI) was (35 ± 19)%; control group (16 ± 8%). The expression of p16 protein in the chemotherapy group was significantly higher than that in the control group (P < 0.01). The expression of Rb protein in cancer cells after preoperative regional arterial infusion chemotherapy was (4 8 ± 11) %, which was significantly higher than that in the control group (LI was (38 ± 10) %), P <0 01. The expression of p16 protein and Rb protein was significantly associated with Dukes stage, with a higher expression in stage B than in stages C and D. Conclusion Preoperative regional arterial infusion chemotherapy can up-regulate the expression of p16 and Rb proteins and inhibit the cell cycle from entering into S phase at the G1/S restriction point.
