目的:培养人眼睫状肌细胞,研究其生长、超微结构、免疫组织化学特性和功能特点。方法:采用组织块培养法对10只青年供体眼睫状肌进行培养,观察其生长特性,用透射电子显微镜检查其超微结构,进行结蛋白免疫组织化学检查,应用单细胞收缩分析技术研究卡巴可对细胞收缩的影响。结果:从人眼睫状肌组织培养了细胞并传代,细胞融合后呈峰谷样排列;透射电子显微镜下可见丰富的微丝;培养的细胞结蛋白染色阳性,组织切片中睫状肌细胞结蛋白染色阳性,成纤维细胞,血管内皮细胞结蛋白染色阴性,小血管壁呈弱到中度阳性。10~(-3)M卡巴可可以引起培养的细胞收缩,此作用被10~(-3)M阿托品拮抗。结论:培养的细胞符合人眼睫状肌细胞特征,具有收缩功能。眼科学报1998;14:69—72。
Objective: To cultivate ciliary muscle cells of human eye and study its growth, ultrastructure, immunohistochemistry and functional characteristics. Methods: Tissue explant culture was used to culture ciliary muscles of 10 young donors. The growth characteristics were observed. The ultrastructure of the ciliary muscles was observed by transmission electron microscopy. The immunohistochemical examination of the protein was performed. The single cell contraction analysis Kabbah cells on the impact of contraction. Results: The cells were cultured and subcultured from ciliary muscle of human eye, and the cells were arranged in a peak-valley arrangement after fusion. Transmission electron microscopy showed abundant microfilaments. The cultured cells were positive for histone densin staining and ciliary muscle cell junction Positive protein staining, fibroblasts, endothelial cell protein-negative staining, small blood vessel wall was weak to moderately positive. The effect of 10 ~ (-3) M carbachol on contractile activity of cultured cells was antagonized by 10 ~ (-3) M atropine. Conclusion: The cultured cells conform to the characteristics of human ciliary muscle cells and have systolic function. Ophthalmic Science 1998; 14: 69-72.
