目的分别采用两种荧光染料对人体蠕形螨染色后置于激光共聚焦显微镜下进行三维重建,对其重要结构进行功能性探讨。方法先用异硫氰酸荧光素(fluorescein isothiocyanate,FITC)对虫体进性初染,再用碘化丙啶(propidium iodide,PI)复染,置于激光共聚焦显微镜下,用Imaris5.0软件进行三维重建,观测、鉴定毛囊蠕形螨形态特征。结果异硫氰酸和碘化丙啶均能与虫体充分结合,在三维重建技术下清楚观察到了虫体外部形态及前段摄食通道等内部结构。结论荧光双染和三维重建技术的结合,对深入研究人体蠕形外部形态、内部结构及探讨其功能、意义奠定了坚实基础。
OBJECTIVE: Two kinds of fluorescent dyes were used to stain human Demodex and then placed under confocal laser scanning microscopy for three-dimensional reconstruction. Functional studies were carried out on their important structures. Methods The cells were initially infected with fluorescein isothiocyanate (FITC) and then stained with propidium iodide (PI). The cells were harvested under Confocal laser scanning microscope Software three-dimensional reconstruction, observation, identification Morphological characteristics Demodex folliculorum. Results Both isothiocyanate and propidium iodide could be fully integrated with parasites. The internal structure of the parasites and their feeding channels were clearly observed under the three-dimensional reconstruction technique. Conclusion The combination of fluorescence double staining and three-dimensional reconstruction technology has laid a solid foundation for further study on the external appearance and internal structure of human body and its function and significance.
