目的:通过观察益肺汤对人肺腺癌耐顺铂细胞株(A549/DDP)移植瘤裸鼠脾脏指数和肺癌多药耐药P-糖蛋白(Pgp)基因表达的影响,探讨益肺汤对裸鼠免疫功能的影响和逆转肺癌多药耐药的作用机制。方法:建立A549/DDP裸鼠移植瘤模型,随机分为对照组、顺铂组、益肺汤低剂量组、益肺汤中剂量组、益肺汤高剂量组、益肺汤中剂量+顺铂组,共6组,每组10只,分别给与相应的药物干预。给药21天后,处死小鼠取瘤组织和脾脏,称脾重,计算各组脾脏指数;检测瘤组织基因P-gp的表达情况。结果:与对照组比较,顺铂组的脾脏指数显著降低,益肺汤中、高剂量组的脾脏指数有所提高,差异均有统计学意义(P<0.05);与顺铂组比较,益肺汤低、中、高各剂量组及益肺汤中剂量+顺铂组的脾脏指数均显著提高,差异均有统计学意义(P<0.05)。益肺汤低、中、高各剂量组及益肺汤中剂量+顺铂组的P-gp基因的表达量均低于对照组,而顺铂组的表达量高于对照组,差异均有统计学意义(P<0.05)。结论:益肺汤能提高裸鼠的免疫功能,逆转非小细胞肺癌多药耐药,其逆转机制是通过下调多药耐药基因P-gp的表达来实现的。
OBJECTIVE: To observe the effects of Yifei Decoction on spleen index and multidrug resistance P-glycoprotein (Pgp) gene expression in human lung adenocarcinoma A549 / DDP xenografts in nude mice, On immune function of nude mice and reversing the mechanism of multidrug resistance in lung cancer. Methods: The A549 / DDP xenografted nude mice models were established and randomly divided into control group, cisplatin group, Yifei Decoction low dose group, Yifei Decoction middle dose group, Yifei Decoction high dose group, Yifei Decoction plus cis Platinum group, a total of 6 groups, 10 in each group, were given the appropriate drug intervention. Twenty-one days after the administration, the mice were sacrificed to take the tumor tissue and the spleen, and the spleen weights were calculated. The spleen index of each group was calculated. The expression of P-gp in the tumor tissue was detected. Results: Compared with the control group, the spleen index of cisplatin group was significantly decreased, while the spleen index of Yifei decoction group was higher than that of cisplatin group (P <0.05). Compared with cisplatin group, The spleen indexes of low, middle and high doses of lung soup, medium dose of Yifei decoction + cisplatin group were significantly increased, the differences were statistically significant (P <0.05). Yifei Decoction low, middle and high dose groups and Yifei Tang medium dose + cisplatin group P-gp gene expression levels were lower than the control group, while the expression of cisplatin group was higher than the control group, the difference was Statistical significance (P <0.05). Conclusion: Yifei Decoction can improve the immune function of nude mice and reverse the multidrug resistance in non-small cell lung cancer. The mechanism of YIP decoction is to down-regulate the expression of multidrug resistance gene P-gp.
