目的:建立测定人血浆中硫酸头孢匹罗浓度的高效液相色谱方法。方法:采用100μL7%高氯酸沉淀200μL血浆样品蛋白,高速离心后取上清液20μL进样分析。以Kromasil C18柱(4.6mm×250mm,5μm)为分析柱,50mmol.L-1磷酸二氢钠溶液-乙腈(87∶13)为流动相,流速为1.0mL.min-1,检测波长为270nm,盐酸可乐定为内标物。结果:硫酸头孢匹罗在0.312~160.0mg.L-1检测范围内呈良好线性关系(r=0.999 8),平均相对回收率为(102.0±1.2)%,平均绝对回收率为(75.1±1.3)%;日内差异≤3.3%,日间差异≤1.9%,稳定性良好。结论:该方法操作简便、灵敏、准确、稳定,适用于血浆硫酸头孢匹罗的测定。
Objective: To establish a method for the determination of cefpiiro sulphate in human plasma by high performance liquid chromatography. Methods: 200μL of plasma protein was precipitated with 100μL of 7% perchloric acid. After high speed centrifugation, 20μL of supernatant was injected for analysis. The mobile phase was a Kromasil C18 column (4.6 mm × 250 mm, 5 μm) with a mobile phase of 50 mmol·L -1 sodium dihydrogenphosphate solution (87:13) at a flow rate of 1.0 mL · min -1 with a detection wavelength of 270 nm , Colaidine hydrochloride as an internal standard. Results: Cefpiiro sulphate showed a good linearity (r = 0.999 8) within the range of 0.312-160.0 mg.L-1, the average relative recovery was (102.0 ± 1.2)% and the average absolute recovery was (75.1 ± 1.3) )%; Intraday difference ≤3.3%, daytime difference ≤1.9%, good stability. Conclusion: The method is simple, sensitive, accurate and stable and is suitable for the determination of cefpirome sulfate in plasma.
