目的测试不同浓度的重组人肿瘤坏死因子(rHTNF)及五种化疗药物联合后,对人卵巢癌细胞的毒性作用。方法应用噻唑蓝法(MTT)观察药物对体外培养的人卵巢癌细胞系SKOV3的毒性。结果rHTNF低浓度与5-Fu、CP的各浓度及CDDP中、低浓度联合呈相加作用,与CDDP高浓度联合呈协同作用,与CTX、TSPA各浓度联合呈拮抗作用。rHTNF高浓度与5-Fu、CDDP、CP各浓度联合呈协同作用,与TSPA各浓度联合呈相加作用,与CTX各浓度联合呈拮抗作用。结论TNF与化疗药物联合后能否出现协同作用,不仅与药物性质有关,还与药物浓度有关,必须在一定浓度下,才有协同作用
Objective To investigate the toxic effects of different concentrations of recombinant human tumor necrosis factor (rHTNF) and five chemotherapeutic agents on human ovarian cancer cells. Methods MTT assay was used to observe the toxicity of the drug on human ovarian cancer cell line SKOV3 cultured in vitro. Results Low concentrations of rHTNF combined with 5-Fu, CP and low and medium concentrations of CDDP showed a synergistic effect, which was synergistic with high concentrations of CDDP and antagonistic with each concentration of CTX and TSPA. High concentrations of rHTNF and 5-Fu, CDDP, CP concentrations showed a synergistic effect, combined with each concentration of TSPA was additive, and CTX concentrations were antagonistic. Conclusions The synergistic effect between TNF and chemotherapeutic drugs can not only be related to the nature of the drug but also related to the concentration of the drug.
