以10个向日葵品种为试材,利用细胞悬浮技术,研究了培养耐盐细胞系的最佳条件。结果表明:切取向日葵7日龄无菌苗子叶与胚轴接种于6-BA 0.5mg·L~(-1)+NAA 0.5mg·L~(-1)+NaCl 50mmol·L~(-1)+蔗糖40g·L~(-1),pH 5.5~7.0的MS培养基中,培养温度为28℃,14d后取其1g愈伤组织接种于添加NaCl 175mmol·L~(-1)的30mL液体培养基中,振荡培养,7d继代1次,即可获得稳定耐盐细胞系。该方案不仅适合油葵也适用食葵。
Ten sunflower cultivars were used as materials to study the best conditions for cultivating salt-tolerant cell lines by using cell suspension technology. The results showed that the leaves and hypocotyls of 7-day-old germ-free seedlings were cut into 6-BA 0.5 mg · L -1 NAA 0.5 mg · L -1 NaCl 50 mmol·L -1, + Sucrose 40 g · L -1 and pH 5.5-7.0. The culture temperature was 28 ℃. After 14 days, 1 g callus was inoculated into 30 mL of liquid containing 175 mmol·L -1 NaCl Medium, shaking culture, 7d subculture 1 time, you can get a stable salt-tolerant cell line. The program is not only suitable for sunflower oil sunflower also apply.
