目的:观察吗啡对体外培养正常人体子宫平滑肌细胞增殖的影响。方法:制备不同浓度的吗啡及其拮抗剂纳络酮,作用于培养的子宫平滑肌细胞上,通过四甲基偶氮唑盐比色法(MTT法)检测吗啡对其的影响。结果:不同浓度的吗啡组与对照组测定的490nm吸光值(OD490值)相比,P<0.05;1×10-7%、1×10-6%、1×10-5%和1×10-4%吗啡组的细胞增殖速度依次降低;而吗啡(1×10-5%)+纳络酮(1×10-5%)组与对照组OD490值相比差异不明显。结论:外源性阿片类药物吗啡可直接抑制人体子宫平滑肌细胞的增殖。
Objective: To observe the effect of morphine on the proliferation of normal human uterine smooth muscle cells in vitro. Methods: Different concentrations of morphine and its antagonist naloxone were prepared and cultured on cultured uterine smooth muscle cells. The effects of morphine on them were detected by MTT assay. Results: Compared with 490 nm absorbance (OD490 value) of morphine group at different concentrations, the difference was statistically significant (P <0.05); 1 × 10-7%, 1 × 10-6%, 1 × 10-5% and 1 × 10 -4% morphine group decreased in turn the rate of cell proliferation; morphine (1 × 10-5%) + naloxone (1 × 10-5%) group compared with the control group OD490 value was not significantly different. Conclusion: Exogenous opioid morphine can directly inhibit the proliferation of human uterine smooth muscle cells.
