目的:建立夏枯草中异迷迭香酸苷的TLC鉴别和HPLC含量测定方法,为完善夏枯草质量标准提供参考。方法:采用薄层色谱法对夏枯草及夏枯草不同部位中的异迷迭香酸苷进行定性鉴别;采用高效液相色谱法对夏枯草中异迷迭香酸苷进行含量测定,以Agilent Eclipse XDB-C18(4.6 mm×250 mm,5μm)为色谱柱,流速1.0 mL.min-1,检测波长319 nm,柱温30℃,进样体积10μL,流动相16%乙腈-84%水(含1.0%醋酸)等度洗脱。结果:异迷迭香酸苷斑点清晰,且仅存在药用部位果穗中,无干扰;异迷迭香酸苷进样在0.032 667~0.653 34μg与峰面积线性关系良好(r=0.999 9),平均回收率为98.18%,RSD 0.25%,异迷迭香酸苷在夏枯草中的含量>0.1‰。结论:所选指标为夏枯草药用部位果穗中的特征成分,所建立方法操作简单、专属性和重复性良好,可作为夏枯草的质量控制方法。
OBJECTIVE: To establish a method for the TLC identification and determination of HPLC of Promethazine in Prunella vulgaris, and provide a reference for improving the quality standard of Prunella vulgaris. Methods: The TLCs of Prunella vulgaris and Prunella different parts of the different rosmarinic acid were identified qualitatively, HPLC was used to determine the content of Prunella vulgaris, with Agilent Eclipse XDB-C18 (4.6 mm × 250 mm, 5 μm) was used as the chromatographic column at a flow rate of 1.0 mL · min-1 with a detection wavelength of 319 nm and a column temperature of 30 ° C. The injection volume was 10 μL and the mobile phase consisted of 16% acetonitrile to 84% 1.0% acetic acid) isocratic elution. Results: The iso-rosmarin spots were clear, and there was no interference in the ear of the medicinal part. The iso-rosmarin injection had good linearity (r = 0.999 9) between 0.032 667 ~ 0.653 34μg and peak area, The average recovery rate was 98.18%, RSD 0.25%, and the content of isocharisin in Prunella vulgaris was> 0.1 ‰. Conclusion: The selected index is the characteristic component in the ear of Prunella vulgaris. The established method is simple, specific and reproducible, which can be used as the quality control method of Prunella vulgaris.
