目的探讨无并发症的2型糖尿病(T2DM)患者外周血单个核细胞(PBMC)中核因子-κB/p65(NF-κB/p65)、核因子-κB抑制蛋白α(IκB-α)表达水平的变化及其与氧化应激、糖代谢等因素的关系。方法以无并发症T2DM患者30例、非糖尿病对照(NDM)30例为研究对象,测定其PBMC中NF-κB/p65、IκB-α蛋白表达水平,血清丙二醛(MDA)、超氧化物歧化酶(SOD)及糖化血红蛋白(GHb)等指标变化。结果与NDM组相比,T2DM患者NF-κB/p65表达升高;MDA水平升高,SOD活性降低。NF-κB/p65表达与MDA、GHb、稳态模型胰岛素抵抗指数呈正相关,与SOD呈负相关。T2DM患者与NDM间IκB-α蛋白表达无统计学差异。结论无并发症的T2DM患者PBMC中NF-κB/p65表达升高,其表达水平与氧化应激、血糖控制水平相关。
Objective To investigate the expression of NF-κB / p65 and IκB-α in peripheral blood mononuclear cells (PBMC) of patients with type 2 diabetes mellitus (T2DM) without complications Changes and its relationship with oxidative stress, glucose metabolism and other factors. Methods Thirty patients with T2DM without complications and 30 non-diabetic controls (NDM) were enrolled in this study. The expression of NF-κB / p65 and IκB-α in PBMCs were measured. The levels of serum malondialdehyde (MDA), superoxide Dismutase (SOD) and glycosylated hemoglobin (GHb) and other indicators change. Results Compared with NDM group, the expression of NF-κB / p65 was increased in T2DM patients. MDA level and SOD activity were decreased. The expression of NF-κB / p65 was positively correlated with MDA, GHb and homeostasis model insulin resistance index, but negatively correlated with SOD. There was no significant difference in IκB-α protein expression between T2DM patients and NDM patients. Conclusion The expression of NF-κB / p65 in PBMC of non-complication T2DM patients is increased, which is correlated with the level of oxidative stress and blood glucose control.
