目的通过观察灯盏花素在不同时间点对HepG2肝细胞内过氧化物酶体增殖物激活受体-α(PPAR-α)、载脂蛋白A5(apoA5)表达和三酰甘油(TG)含量的影响,旨在初步探讨灯盏花素与PPAR-α、apoA5、TG之间的时间依赖关系,为进一步探索灯盏花素调节TG代谢的具体机制奠定一定的基础。方法在前期实验基础上选取100mmol/L灯盏花素,处理HepG2细胞不同时间点:(1)0h(空白对照)组;(2)6h组;(3)12h组;(4)24h组;(5)36h组;(6)48h组;检测各组细胞内PPAR-α、apoA5基因、蛋白表达水平和TG含量。结果灯盏花素可升高HepG2细胞内PPAR-α、apoA5表达,降低TG含量(P<0.05),并呈时间依赖性。结论灯盏花素能降低肝细胞内TG含量,其机制可能通过使PPAR-α表达水平增加,从而增加apoA5表达水平而实现的。
Objective To observe the effects of breviscapine on the expression of peroxisome proliferator activated receptor-α (PPAR-α), apolipoprotein A5 (apoA5) and triglyceride (TG) in HepG2 hepatocytes at different time points Aiming to investigate the time-dependent relationship between breviscapine and PPAR-α, apoA5 and TG, and lay a foundation for further exploration of the specific mechanism of Breviscapine regulating TG metabolism. Methods 100 mmol / L breviscapine was chosen to treat HepG2 cells at different time points: (1) 0h (blank control group); (2) 6h group; (3) 12h group; (4) 24h group; 5) 36h group; (6) 48h group; the intracellular PPAR-α, apoA5 gene, protein expression level and TG content were detected. Results Breviscapine could increase the expression of PPAR-α, apoA5 in HepG2 cells and decrease the content of TG (P <0.05) in a time-dependent manner. Conclusion Breviscapine can decrease the content of TG in hepatocytes, and its mechanism may be through increasing the expression of PPAR-α and increasing the expression of apoA5.
