猕猴桃营养全面、丰富,含有10多种氨基酸、矿物质、胡萝卜素和多种维生素,对保持人体健康,防病治病具有重要的作用。维生素C又称L-抗坏血酸(ascorbic acid),是含有6碳的不饱和多羟化合物,在医药及营养保健方面的用途十分广泛。维生素C的分析方法有多种,但这些方法往往操作复杂费时,在样品处理过程中很容易使维生素C损失,且灵敏度和精确度不高;而高效液相色谱法(HPLC)具有较高的准确度、精密度和灵敏度,简便快速,更加适用于维生素C的测定。本研究采用高效液相色谱法(HPLC),探究猕猴桃中维生素C的最佳分析条件。通过大量的实验研究,表明以甲醇:纯水=2:8为流动相,流动相的速度为1.2ml/min,紫外检测波长为254nm,柱温在25℃为猕猴桃中维生素C的最佳HPLC分析条件。在此条件下分析猕猴桃中维生素C的含量、稳定性、回收率,所建立的标准曲线具有良好的线性关系,其相关系数R~2=0.9997;其平均回收率为100.21%,相对标准偏差小于1.47%;测得猕猴桃中维生素C的含量为107.33 mg/100g。所建立的方法简单、快速、准确度高,适宜猕猴桃中维生素C含量的分析研究。

Vitamin C, also known as L-ascorbic acid, is a 6-carbon unsaturated polyhydroxy compound that is used in a wide range of medical and nutraceutical uses. There are many ways to analyze vitamin C, but these methods are often complicated and time-consuming. Vitamin C can be easily lost in the sample preparation process with low sensitivity and accuracy. High performance liquid chromatography (HPLC) Accuracy, precision and sensitivity, simple and quick, more suitable for the determination of vitamin C. In this study, high performance liquid chromatography (HPLC) was used to explore the optimum conditions for vitamin C in kiwifruit. Through a large number of experimental studies, the mobile phase was methanol: pure water = 2: 8, the mobile phase was 1.2ml / min, the UV detection wavelength was 254nm, and the optimum temperature was 25 ℃ for the optimal HPLC of vitamin C in kiwifruit Analysis conditions. Under this condition, the content of vitamin C in kiwifruit, the stability and the recovery rate of kiwi fruit were analyzed. The established standard curve had a good linear relationship with the correlation coefficient R ~ 2 = 0.9997; the average recovery was 100.21% and the relative standard deviation was less than 1.47%. The content of vitamin C in kiwifruit was 107.33 mg / 100g. The established method is simple, rapid, high accuracy, suitable for analysis of vitamin C content in kiwifruit.
