目的 观察丹参对慢性低氧性肺动脉高压大鼠肺组织碱性成纤维细胞生长因子(bFGF)表达的影响,探讨丹参阻抑肺腺泡内动脉(IAPA)重建的机制。方法 Wistar大鼠随机分为3组:正常对照组、慢性低氧组、低氧丹参治疗组。低氧大鼠置于常压低氧舱3周后,测定血流动力学;观察IAPA结构变化,用免疫组化检测肺组织bFGF表达;用氯胺T比色法测定肺组织羟田氨酸含量。结果 丹参能降低平均肺动脉压,减少肺组织羟脯氨酸含量(P<0.05,P<0.01),抑制IAPA平滑肌细胞增生,降低肺组织bFGF表达强度 结论(1)丹参能抑制IAPA平滑肌细胞增生及胶原沉积而阻抑IAPA的结构重建。(2)抑制bFGF表达是丹参调节IAPA平滑肌细胞增生的重要途径之一。
Objective To observe the effect of salvia miltiorrhiza on the expression of basic fibroblast growth factor (bFGF) in lung tissue of rats with chronic hypoxic pulmonary hypertension, and to explore the mechanism of Salvia miltiorrhiza inhibiting the reconstruction of intra-aortic internal iliac artery (IAPA). Methods Wistar rats were randomly divided into 3 groups: normal control group, chronic hypoxia group, and hypoxic danshen treatment group. Hypoxic rats were placed in a normobaric hypoxic chamber for 3 weeks. Hemodynamics were measured. The structural changes of IAPA were observed. The expression of bFGF was detected by immunohistochemistry. The hydroxyanthrine was determined by chloramine T colorimetry. content. Results Salvia miltiorrhiza can reduce mean pulmonary arterial pressure, decrease lung hydroxyproline content (P<0.05, P<0.01), inhibit IAPA smooth muscle cell proliferation, and decrease the expression of bFGF in lung tissue. Conclusion (1) Salvia miltiorrhiza can inhibit the proliferation of IAPA smooth muscle cells and Collagen deposition inhibits the structural remodelling of IAPA. (2) Inhibition of bFGF expression is one of the important ways that Salvia Miltiorrhiza regulates the proliferation of smooth muscle cells of IAPA.
