目的:探讨丹参酮ⅡA磺酸钠(STS)对2型糖尿病大鼠心功能的影响。方法:将大鼠随机分为正常(NC)组、模型对照(DM)组及丹参酮ⅡA磺酸钠(DMS)组。成模后DMS组给药12周,留取血标本检查空腹血糖(FPG)、总胆固醇(TC)、甘油三酯(TG)及血浆胰岛素(Ins)水平,麻醉后经颈动脉插管观察心功能的变化,用HE染色观察心肌形态变化。结果:DM组大鼠FPG、血浆Ins、TG及TC水平均高于NC组(P<0.01);DM组与DMS比较,各指标均有下降,但差异均无显著性意义(P>0.05)。DM组大鼠12周时心脏最大舒张期压力变化速率(-dp/dtmax)、最大收缩期压力变化速率(+dp/dtmax)和左室射血分数(LVEF)均降低,与NC组比较,差异均有非常显著性意义(P<0.01);而DMS组经干预治疗12周后-dp/dtmax、+dp/dtmax及LVEF均有上升,与DM组比较,差异均有显著性意义(P<0.05)。DM组大鼠心肌疏松、淡染,心肌纤维肿胀、排列紊乱、空泡变性、有炎症细胞浸润,DMS组心肌细胞排列趋于整齐,结构接近正常。结论:丹参酮ⅡA磺酸钠能够减轻心肌损伤,改善心功能,对糖尿病大鼠的心肌起到保护性作用。
Objective: To investigate the effect of sodium tanshinone Ⅱ A sulfonate (STS) on cardiac function in type 2 diabetic rats. Methods: The rats were randomly divided into normal group (NC), model group (DM) and sodium tanshinone Ⅱ A (DMS) group. After modeling, the rats in DMS group were given 12 weeks of treatment. Blood samples were taken for fasting blood glucose (FPG), total cholesterol (TC), triglyceride (TG) and plasma insulin (Ins) Function changes, with HE staining observed myocardial morphological changes. Results: The levels of FPG, plasma Ins, TG and TC in DM group were higher than those in NC group (P <0.01). There was no significant difference between DM group and DMS (P> 0.05) . The changes of maximal diastolic pressure (-dp / dtmax), maximal systolic pressure changes (+ dp / dtmax) and left ventricular ejection fraction (LVEF) in DM group decreased at 12 weeks. Compared with NC group, (P <0.01). The levels of -dp / dtmax, + dp / dtmax and LVEF increased in DMS group after 12 weeks of intervention, which were significantly different from those in DM group (P <0.05). Myocardium in DM group was loose, lightly stained, swollen and disordered myocardial fibers, vacuolar degeneration and inflammatory cell infiltration. The arrangement of myocardial cells in DMS group tended to be neat and the structure was close to normal. Conclusion: Sodium tanshinone Ⅱ A sulfonate can relieve myocardial injury, improve cardiac function and play a protective role in the myocardium of diabetic rats.
