目的 探讨急性脑缺血后脑组织内磷脂酶A2 (PLA2 )激活及细胞内 [Ca2 + ]i与脑损伤的关系 ,为预防和治疗急性缺血性脑损伤提供理论基础和新的思路。方法 将局灶性脑缺血模型大鼠分 5组(假手术组、缺血 30、 6 0、 90、 12 0min组 ) ,测定脑组织PLA2 活力、脑细胞 [Ca2 + ]i、脑含水量及缺血12 0min组脑组织PLA2 表达量的改变。结果 脑缺血 12 0min脑组织PLA2 活性、 [Ca2 + ]i、脑含水量较假手术组明显升高 ,并与时间呈正相关 ,缺血 12 0min后脑组织中出现sPLA2 ⅡAmRNA表达 ,且cPLA2 ⅣmRNA表达水平较假手术组明显增强。结论 磷脂酶A2 激活参与了脑缺血后神经细胞内钙超载及脑损伤的整个病理过程
Objective To investigate the relationship between phospholipase A2 (PLA2) activation and intracellular [Ca2 +] i and brain injury after acute cerebral ischemia in rats and to provide theoretical basis and new ideas for the prevention and treatment of acute ischemic brain injury. Methods The focal cerebral ischemia model rats were divided into 5 groups (sham operation group, ischemia 30, 60, 90, 12 0min group), the activity of PLA2, brain cell [Ca2 +] i, And the change of PLA2 expression in brain tissue after ischemia for 12 minutes. Results Brain PLA2 activity, [Ca2 +] i and brain water content increased significantly at 12 min after cerebral ischemia compared with sham-operation group, and were positively correlated with time. The expression of sPLA2 ⅡA mRNA in brain tissue and the expression of cPLA2 Ⅳ mRNA Compared with the sham group, the level was significantly increased. Conclusion Phospholipase A2 activation is involved in the whole pathological process of intracellular calcium overload and brain injury after cerebral ischemia
