目的:探讨LINC00958在卵巢癌细胞中对增殖和侵袭中的作用。方法:利用在线软件GEPIA分析TCGA数据库中LINC00958在多种肿瘤组织中的表达;采用qRT-PCR检测转染过表达载体和siRNA后LINC00958的表达水平转染效率;应用CCK8法检测LINC00958对卵巢癌细胞SKOV3和OVCAR3增殖能力的影响;采用Transwell实验检测LINC00958对卵巢癌细胞侵袭能力的影响。结果:经GEPIA软件分析TCGA数据库发现LINC00958在膀胱尿路上皮癌、宫颈鳞癌、头颈部鳞状细胞癌、肺鳞状细胞癌、卵巢浆液性囊腺癌、甲状腺癌、子宫内膜癌和子宫肉瘤等8种肿瘤中表达增加,在急性髓系白血病、皮肤恶性黑色素瘤和睾丸精原细胞肿瘤等3种肿瘤中表达下降;过表达LINC00958后,可增加卵巢癌细胞的增殖和侵袭能力(n P<0.05),而抑制LINC00958的表达可降低卵巢癌细胞的增殖和侵袭(n P<0.05)。n 结论:LINC00958可促进卵巢癌细胞的增殖和侵袭。“,”Objective:To investigate the effect of LINC00958 on proliferation and invasiveness of ovarian cancer cells.Methods:Online software GEPIA was used to analyze the expression of LINC00958 in a variety of tumor tissues archived in the TCGA database. qRT-PCR was used to detect the expression level of LINC00958 as the efficiency of transfection with overexpression vector and siRNA. CCK8 method was used to detect the effect of LINC00958 on the proliferation of ovarian cancer SKOV3 and OVCAR3 cells. Transwell assay was used to detect the effect of LINC00958 on the invasiveness of ovarian cancer cells.Results:GEPIA analysis of TCGA database found that LINC00958 expression was up-regulated in bladder urothelial carcinoma, cervical squamous cell carcinoma, head and neck squamous cell carcinoma, lung squamous cell carcinoma, ovarian serous cystadenocarcinoma, thyroid cancer, endometrial cancer and uterine cancer, and was down-regulated acute myeloid leukemia, malignant melanoma of the skin and testicular seminoma. Overexpression of LINC00958 could increase the proliferation and invasiveness of ovarian cancer cells (n P<0.05) , while inhibition of the LINC00958 expression act the opposite (n P<0.05) .n Conclusion:LINC00958 may promote the proliferation and invasiveness of ovarian cancer cells.
