作为高效液相色谱使用的伏安检测器是在带有管状铂工作电极的三电极系统基础上发展起来的。它的死体积小(约1 μl),可在直流极谱法和示差脉冲极谱法这两种方式下应用。它的性能已用苯酚、抗坏血酸和尿酸等各种有机物作了试验。检测限约为0.5ng,线性动态范围至少有五个浓度数量级,重现性好。直流极谱法和示差脉冲极谱法具有大致相同的检测灵敏度和类似的峰面积重现性。但是,直流极谱法的峰高比示差脉冲极谱法的峰高重现性更好。在反相系统中,该检测器可以把儿茶酚胺类的肾上腺硷、正肾上腺硷、多巴胺和L—多巴分离开。能检测至毫微克量级,峰形窄而且对称。
The voltammetric detector used as a HPLC has evolved from the three-electrode system with tubular platinum working electrode. Its small dead volume (about 1 μl) can be used in both polarography and differential polarography. Its performance has been tested with various organic compounds such as phenol, ascorbic acid and uric acid. The detection limit is about 0.5ng, and the linear dynamic range is at least five orders of magnitude, good reproducibility. DC polarography and differential pulse polarography have approximately the same detection sensitivity and similar peak area reproducibility. However, the peak height of direct current polarography is better than that of differential pulse polarography. In an inverting system, the detector separates the epinephrine, norepinephrine, dopamine and L-dopa of catecholamines. Can detect to the nanogram level, narrow and symmetrical peak shape.
