目的探讨蛋白多糖Decorin对博莱霉素(BLM)所致大鼠肺间质纤维化过程中的转化生长因子-β1(TGF-β1)的影响及其治疗作用。方法90只Wistar大鼠随机分为3组:对照组(A)、模型组(B)、治疗组(C)。对照组气管内注入生理盐水(作为阴性对照),模型组经气管内注入BLM复制肺纤维化模型,治疗组大鼠于BLM灌注后每日腹腔内注入Decorin(质量浓度0·1g/L,每只大鼠注入0·5mL)进行干预。各组动物均于气管内灌注后第7、14、28天分别处死10只,取肺组织做TGF-β1的免疫组化并行图像分析、羟脯氨酸含量测定,HE染色。结果治疗组与模型组比较TGF-β1表达明显减低(P<0·05);肺泡炎、肺纤维化明显减轻;羟脯氨酸含量明显减低。结论Decorin对BLM所引起的肺纤维化具有明显的治疗作用。
Objective To investigate the effect of proteoglycan Decorin on transforming growth factor-β1 (TGF-β1) during the process of pulmonary interstitial fibrosis induced by bleomycin (BLM) in rats and its therapeutic effect. Methods Ninety Wistar rats were randomly divided into three groups: control group (A), model group (B) and treatment group (C). In the control group, physiological saline was injected into the trachea (negative control). The model group was injected intratracheally with BLM to induce the pulmonary fibrosis model. The rats in the treatment group were given daily intraperitoneal injection of Decorin (mass concentration 0.1 g / L, Only rats were injected with 0.5 mL). Each group of animals were sacrificed on the 7th, 14th and 28th days after intratracheal instillation. Ten rabbits were sacrificed and lung tissue was harvested for immunohistochemical analysis of TGF-β1, hydroxyproline content and HE staining. Results Compared with the model group, the expression of TGF-β1 in the treatment group was significantly decreased (P <0.05); the alveolitis and pulmonary fibrosis were significantly reduced; and the hydroxyproline content was significantly decreased. Conclusion Decorin has a significant therapeutic effect on pulmonary fibrosis induced by BLM.
