目的:探讨玉米须提取液对糖尿病大鼠肾脏的保护作用及机制研究。方法:采用健康雄性Sqrague-Dawley大鼠随机分为正常对照组、糖尿病组及玉米须治疗组,第4、8周任取6只大鼠,收集血、尿标本测定生化指标。大鼠肾组织分别作组织学、超微结构、免疫组织化学方法检测肾小球podocin蛋白表达,应用逆转录-聚合酶链反应(RT-PCR)观察podocin mRNA在肾组织中的表达。结果:在4周、8周,糖尿病组血糖、肌酐清除率、尿白蛋白排泄率较正常对照组均有不同程度的升高。玉米须提取液治疗组血糖、肌酐清除率、尿白蛋白排泄率较正常对照组均有不同程度的升高。玉米须提取液治疗组血糖、肌酐清除率、尿白蛋白排泄率显著低于同时期的糖尿病组。糖尿病组大鼠肾组织podocin mRNA和蛋白表达较正常对照组明显降低。玉米须提取液组明显上调。结论:玉米须提取液对糖尿病大鼠肾结构功能具有保护作用,可影响肾小球上皮细胞足突podocin蛋白表达,从而对糖尿病肾病有保护作用。
Objective: To study the protective effect and mechanism of corn stubble extract on kidneys of diabetic rats. METHODS: Healthy male Sqrague-Dawley rats were randomly divided into normal control group, diabetic group and corn beard treatment group, and 6 rats were taken at the 4th and 8th weeks. Blood and urine specimens were collected to measure biochemical indicators. Renal tissue of rats was used for histology, ultrastructure, and immunohistochemistry to detect glomerular podocin protein expression. Reverse transcription polymerase chain reaction (RT-PCR) was used to observe the expression of podocin mRNA in renal tissue. RESULTS: At 4 and 8 weeks, the blood glucose, creatinine clearance, and urinary albumin excretion rate in the diabetic group were increased to varying degrees compared with the normal control group. The blood glucose, creatinine clearance, and urinary albumin excretion rate of the corn silk extract treatment group were increased to varying degrees compared with the normal control group. The blood sugar, creatinine clearance, and urinary albumin excretion rate were significantly lower in the treatment group than in the diabetic group at the same time. The expression of podocin mRNA and protein in kidney tissue of diabetic rats was significantly lower than that of normal control group. Corn silk extract group was significantly upregulated. Conclusion: Corn silk extract has a protective effect on renal structural function in diabetic rats, and can affect the expression of podocin protein in the glomerular epithelial foot processes, and thus has a protective effect on diabetic nephropathy.
