目的:研究姜黄素对人乳腺癌MCF-7细胞信号传导通路相关蛋白激酶的影响。方法:以MTT比色法观察不同质量浓度姜黄素及其在不同时间点对MCF-7细胞的增殖抑制作用;western blot法观察磷酸化蛋白激酶p-JNK、p-p38、p-ERK和p-NF-κB的表达。结果:姜黄素干预MCF-7细胞72h后的半数抑制浓度(IC50)为22.48μmol/L,且其增殖抑制效应呈剂量及时间依赖关系。40μmol/L姜黄素抑制了p-ERK、p-NF-κB和p-p38的表达,但没有影响p-JNK的表达。结论:姜黄素能抑制乳腺癌MCF-7细胞的增殖,其机制可能与信号传导通路相关蛋白激酶的活性有关。
Objective: To study the effect of curcumin on the protein kinase involved in signal transduction pathway of human breast cancer MCF-7 cells. Methods: MTT assay was used to observe the effects of different concentrations of curcumin on the proliferation of MCF-7 cells at different time points. The phosphorylation of p-JNK, p-p38, p-ERK and p NF-κB expression. Results: The median inhibitory concentration (IC50) of curcumin in MCF-7 cells after 72 hours was 22.48μmol / L, and its proliferation inhibitory effect in a dose-and time-dependent manner. 40μmol / L curcumin inhibited the expression of p-ERK, p-NF-κB and p-p38, but did not affect the expression of p-JNK. Conclusion: Curcumin can inhibit the proliferation of breast cancer MCF-7 cells, and its mechanism may be related to the activity of signal transduction pathway-related protein kinases.
