目的建立同时测定野菊花中绿原酸、咖啡酸和蒙花苷含量的方法。方法采用高效液相色谱法,应用Agilent Eclipse C18柱(250 mm×4.6 mm,5μm),二极管阵列检测器(DAD),以甲醇-乙腈-0.2%乙酸溶液为流动相,梯度洗脱,流速为1 m L·min?1,柱温为30℃,检测波长为334 nm。结果绿原酸、咖啡酸和蒙花苷的峰面积与浓度的线性关系良好(r>0.999 3),平均加样回收率分别为99.5%,100.4%,100.4%。结论该方法简单、准确、重复性好,可为全面评价和控制野菊花的质量提供依据。
Objective To establish a method for the simultaneous determination of chlorogenic acid, caffeic acid and monatin in wild chrysanthemum. Methods High performance liquid chromatography (HPLC) was used to elute gradient elution with a gradient of elution gradient of methanol-acetonitrile-0.2% acetic acid solution on an Agilent Eclipse C18 column (250 mm × 4.6 mm, 5 μm) and a diode array detector 1 m L · min -1, the column temperature was 30 ℃ and the detection wavelength was 334 nm. Results The linear relationship between the peak area of chlorogenic acid, caffeic acid and mandarin was good (r> 0.999 3). The average recoveries were 99.5%, 100.4% and 100.4%, respectively. Conclusion The method is simple, accurate and reproducible. It can provide a basis for the comprehensive evaluation and control of the quality of wild chrysanthemum.
