目的:以SH-SY5Y细胞作为神经元代表,建立β淀粉样蛋白(Aβ_(25-35))诱导细胞损伤的阿尔茨海默病(Alzheimer’s Disease,AD)体外细胞筛选模型,并将其应用于筛选39个中药材提取物中的杭神经细胞损伤活性组分。方法:采用CCK-8法进行活性初筛,CCK-8法和Hoechst33342/PI双染色分析法复筛活性组分。结果:初筛和复筛结果一致表明,39个药材提取物中黄柏40%乙醇部位(HB40)和95%乙醇部位(HB95)有较好的抗Aβ损伤活性。结论:提示黄柏有潜在的治疗杭阿尔茨海默病的药用价值。
OBJECTIVE: To establish an in vitro cell selection model of Alzheimer’s Disease (AD) induced by β-amyloid protein (Aβ_ (25-35)) using SH-SY5Y cells as neurons, Screening of 39 Chinese herbal extracts of Hangxing cells injury activity components. Methods: CCK-8 method was used to conduct primary screening. CCK-8 and Hoechst33342 / PI double staining were used to screen the active components. Results: The results of preliminary screening and rescreening showed that the antimicrobial activity of 40% ethanol (HB40) and 95% ethanol (HB95) in the cortex phellodendri cv. Conclusion: Cork has potential medicinal value for treatment of Alzheimer’s disease.
