目的 探讨介入热疗对肝组织及肿瘤组织血管渗透性的影响及意义 ,为研究介入热疗的生物学效应机制奠定基础 .方法 建立可供实验研究的兔 VX2 移植性肝癌模型 30只 ,随机分为 3组 (非灌注组、普通灌注组及热灌注组 ,每组n= 10 ) ,在 X线监视下 ,经股动脉插管至肝动脉 .进行灌注 ,灌注液为生理盐水 (温度为 6 0℃ ) ,液量 30 m L ,15 min缓慢推注 .灌注结束前 5 min,经导管推注 10 g· L- 1伊文思蓝(Evans Blue,EB) ,2 m L· kg- 1 ,非灌注组直接推注 EB,灌注EB后置 10 min,取出肝脏用生理盐水灌注肝动脉冲洗血管中残留的 EB.切取靠近肝门部的小块正常肝组织、瘤组织 ,称质量后 ,放入 1m L甲酰胺液中 ,置于 5 0℃恒温水浴箱 6 0 h,提取液用 72 2型光栅分光光度计测出 A6 2 0 nm,从标准曲线上测出相应的 EB含量 ,以反映该组织毛细血管的渗透性 .结果 肿瘤组织与肝组织的 EB含量在 3组中均有差别 (P<0 .0 5 ) ;普通灌注组与非灌注组肝组织的 EB含量、肿瘤组织的 EB含量无显著差别 (P>0 .0 5 ) ;热灌注组与非灌注组、普通灌注组比较 ,肝组织的 EB含量、肿瘤组织的 EB含量高 ,差别显著(P<0 .0 5 ) .结论 介入热疗可以增加肝组织及肿瘤组织的血管渗透性 .
Objective To investigate the effect and significance of interventional hyperthermia on vascular permeability in liver and tumor tissues and lay the foundation for the study of the biological effect mechanism of interventional hyperthermia.Methods Thirty rabbits with experimental VX2 liver transplantation model were randomly divided (Non-perfusion group, normal perfusion group and hot perfusion group, each group n = 10) under the X-ray monitoring, the femoral artery catheterization to the hepatic artery perfusion, perfusion fluid was saline (temperature of 6 0 ℃), the volume of liquid was 30 m L, and the bolus was injected slowly at 15 min.Entaxed 10 g · L-1 Evans Blue (EB), 2 m L · kg-1, The non-perfusion group was injected with EB directly, followed by infusion of EB for 10 min, and then the residual EB in the hepatic artery was removed by perfusion of the hepatic artery with normal saline.A small piece of normal liver tissue and tumor tissue near the hepatic portal were excised, Into 1m L formamide solution, placed in 50 ℃ water bath 60h, extract 72 2 Grating spectrophotometer A620nm, measured from the standard curve of the corresponding EB content to reflect The capillary permeability of the tissue.Results The content of EB in tumor tissue and liver tissue was in 3 groups (P <0.05). There were no significant differences in EB content and EB content in liver between normal perfusion group and non-perfusion group (P> 0.05) Compared with the perfusion group, the EB content in liver tissue and the EB content in tumor tissue were higher (P <0.05) .Conclusion Interventional hyperthermia can increase the vascular permeability of liver tissue and tumor tissue.
