用不同浓度胶原酶分离小鼠腔前卵泡,以4 mg/ ml 胶原酶处理组所获采集量大且对腔前卵泡早期培养无影响。培养液中分别加入20 % 牛卵泡液、FSH+ LH( 各10IU/ ml) 和胰岛素(0 .01ug/ ml) ,结果加入20 % 牛卵泡液组的腔前卵泡在培养10d 后卵泡直径明显大于对照组,而GVBD 发生率明显低于对照组;加入FSH + LH( 各10IU/ ml) 或胰岛素(0 .01ug/ ml) 组的腔前卵泡GVBD 发生率明显高于对照组。提示:FSH 、LH、胰岛素能促进腔前卵泡体外成熟,卵泡液能促进腔前卵泡体外生长而抑制其体外成熟
Pre-parenchymal follicles were isolated from mice with different concentrations of collagenase and collected in 4 mg / ml collagenase group without any effect on early culture of preantral follicles. The follicles of 20% bovine follicular fluid, FSH + LH (each 10IU / ml) and insulin (0. 01ug / ml) were added into the culture medium. The diameter of follicle in 20% Group, while the incidence of GVBD was significantly lower than that of the control group. The incidence of GVBD in the preantral follicles of FSH + LH group (10 IU / ml) or insulin (0. 01 ug / ml) group was significantly higher than that of the control group. Tip: FSH, LH, insulin can promote the premature rupture of follicles in vitro, follicular fluid can promote preantral follicles in vitro growth and inhibit its in vitro maturation
