目的探讨益气清化方对高脂诱导非酒精性脂肪性肝病模型大鼠肠道菌群的影响。方法选用清洁级健康成年Wistar大鼠30只,随机分为空白组、模型组、治疗组,每组10只。模型组及治疗组予高脂饲料制备非酒精性脂肪性肝病模型。治疗组于造模的第1天同时灌胃给予益气清化方,模型组灌服等量的0.9%Na Cl溶液,空白组予普通饲料及等量的0.9%Na Cl溶液。各组均干预8周。实验结束后,测定大鼠肝功能(血清ALT、AST、TG、TP)、血浆内毒素(LPS)水平;采集大鼠肝组织,HE染色后观察肝脂肪变性情况;ERIC-PCR指纹图谱检测肠道菌群情况,并进行相似性聚类分析。结果 1模型组ALT、AST、TG、TC及LPS水平较空白组明显升高(P<0.05);治疗组ALT、AST和LPS水平明显低于模型组(P<0.05)。2治疗组可见肝细胞少量的脂肪细胞浸润,较模型组肝细胞炎症和坏死明显减少。3与模型组比较,治疗组肠道菌群ERIC-PCR指纹图谱的条带数目更多、分布区域更宽,提示治疗组大鼠在益气清化方的干预下其肠道菌群的构成有所恢复。4聚类分析显示,空白组与治疗组肠道菌群的相似性明显大于空白组与模型组以及模型组与治疗组的相似性,提示益气清化方能够纠正非酒精性脂肪性肝病大鼠的肠道菌群紊乱;大鼠肠道菌群多样性分析显示,空白组和治疗组的多样性明显高于模型组,提示益气清化方可增加非酒精性脂肪性肝病大鼠肠道菌群的种类,进而改善肠道菌群紊乱。结论益气清化方能够显著减轻非酒精性脂肪性肝病大鼠肝细胞的损伤和肝脏的脂肪沉积,并有助于改善肠道菌群紊乱。
Objective To investigate the effect of Yiqi Qinghua Recipe on the intestinal microbiota in rats with non-alcoholic fatty liver disease induced by high fat. Methods Thirty healthy adult Wistar rats were randomly divided into blank group, model group and treatment group, with 10 rats in each group. The model group and the treatment group were given a high-fat diet to prepare a nonalcoholic fatty liver disease model. The treatment group was given intragastric administration of Yiqiqinghuafang at the same time on the first day of modeling. The model group was given equal amount of 0.9% NaCl solution, and the blank group was given normal diet and the same amount of 0.9% NaCl solution. Each group was intervened for 8 weeks. After the end of the experiment, the liver function (serum ALT, AST, TG, TP) and plasma endotoxin (LPS) levels were measured; the liver tissue of the rats was harvested, and hepatic steatosis was observed after HE staining; the ERIC-PCR fingerprint was used to detect the intestines. Conclusion Yiqi Qinghua Recipe can significantly reduce liver cell damage and hepatic fat deposition in rats with non-alcoholic fatty liver disease, and it can help to improve intestinal flora disturbance.
