目的:观察PPARγ对人胰腺癌细胞中抑癌基因PTEN表达的影响。方法:采用体外培养人胰腺癌细胞株PANC-1,使用罗格列酮、GW9662细胞处理,运用RT-PCR检测PANC-1细胞中的PTEN基因变化情况。结果:PPARγ激动剂罗格列酮可诱导抑癌基因PTEN的高表达,随着罗格列酮浓度增加,表达也逐渐增强。结论:胰腺癌细胞中PPARγ的表达和抑癌基因PTEN的表达呈一致性,PPARγ可能是通过一定信号的转导通路来调节抑癌基因PTEN的表达,以发挥抗肿瘤的作用。
Objective: To observe the effect of PPARγ on the expression of tumor suppressor gene PTEN in human pancreatic cancer cells. Methods: Human pancreatic cancer cell line PANC-1 was cultured in vitro and treated with rosiglitazone and GW9662 cells. The changes of PTEN gene expression in PANC-1 cells were detected by RT-PCR. RESULTS: PPARγ agonist rosiglitazone induced a high expression of the tumor suppressor gene PTEN, and its expression gradually increased with the increase of rosiglitazone concentration. Conclusion: The expression of PPARγ in pancreatic cancer cells is consistent with the expression of tumor suppressor gene PTEN. PPARγ may play an anti-tumor role by regulating the expression of tumor suppressor gene PTEN through a signal transduction pathway.
