以新铁炮百合叶片、花瓣、花丝、鳞片和种子为外植体,进行组织培养再生研究。结果表明:不同外植体诱导分化的能力不同,其中种子>鳞片>花丝>花瓣>叶片,诱导率分别为98.27%,95.83%,86.50%,14.63%和11.43%。不同继代培养基对新铁炮离休快繁有显著影响,选用MS作为基本培养基,附加不同浓度的BA和NAA配合成10个组合,并进行随机区组实验,结果表明:MS+BA2mg·L-1+NAA0.2mg·L-1继代繁殖效果最佳,繁殖倍数40d达14~17株苗。不同生根培养基对新铁炮百合继代苗生根影响差异显著,其中以MS不附加任何激素为最佳,生根率为99%,平均每株苗生粗根6.9条。不同移栽基质对新铁炮百合生根苗移栽成活率影响不同,其中以细河沙:草炭(2:1)为最佳,移栽成活率可达92%。
Using new Lilium lily leaves, petals, filaments, scales and seeds as explants for tissue culture regeneration research. The results showed that the ability of different explants to differentiate was different. Seed> scale> filament> petal> leaf, the induction rate was 98.27%, 95.83%, 86.50%, 14.63% and 11 respectively. 43%. The effects of different transplanting media on the survival rate of transplanting of Lilium regia Lilium seedlings were different, among which fine sand and peat (2: 1) was the best, and the transplanting survival rate was 92%.
