目的 建立人羊膜 (HAM)负载培养骨髓间充质干细胞 (MSCs)生长的组织工程方法 ,观察MSCs生长增殖的形态特点。方法 将贵州小香猪的MSCs原代培养 ,传代扩增后 ,以不同密度多次接种于HAM基质面 ,逐日于倒置显微镜下观察MSCs的生长增殖变化情况 ,并于培养 8、1 8d后进行光镜、电镜观察。结果 接种后 30min内明显见到MSCs在HAM上贴附生长 ,MSCs能以合适的接种密度在羊膜上良好生长。结论 HAM对MSCs有明显的促增殖作用 ,MSCs可以HAM为载体在体外进行培养 ,HAM是MSCs的良好载体
Objective To establish a tissue engineering method for the growth of human mesenchymal stem cells (MSCs) cultured in human amniotic membrane (HAM) and observe the morphological characteristics of the growth and proliferation of MSCs. Methods The MSCs of Guizhou Xiao Xiang pigs were primary cultured, passaged and expanded, and then inoculated on HAM matrixes at different densities for several times. The growth and proliferation of MSCs were observed daily under inverted microscope and cultured for 8 and 18 days Light microscopy, electron microscopy. Results MSCs apparently grew on HAM within 30 min after inoculation, and MSCs grew well on amniotic membrane with suitable inoculation density. Conclusion HAM can significantly promote the proliferation of MSCs. MSCs can be cultured in vitro using HAM as a carrier. HAM is a good carrier of MSCs
