本实验探讨了TNF-α和IFN-γ对人胰腺癌SW-1990细胞中CD44标准型及变异型蛋白表达的调节作用。分别用250U/ml的TNF-α和IFN-γ处理培养中的人胰腺癌细胞株SW-1990,流式细胞和免疫组化方法检测显示,250U/ml的TNF-α和IFN-γ均可上调SW-1990细胞中CD44变异型蛋白的表达。提示细胞因子的作用是人胰腺癌细胞CD44蛋白表达变化的可能机制之一;TNF-α和IFN-γ可能通过上调肿瘤细胞CD44变异型蛋白的表达,而参与了肿瘤的浸润、转移过程。
This experiment explored the regulatory effects of TNF-α and IFN-γ on the expression of CD44 standard and variant proteins in human pancreatic cancer SW-1990 cells. Human pancreatic cancer cell line SW-1990 was treated with 250 U/ml TNF-α and IFN-γ, respectively. Flow cytometry and immunohistochemistry showed that 250 U/ml TNF-α and IFN-γ were Upregulate the expression of CD44 variant protein in SW-1990 cells. It suggests that the role of cytokines is one of the possible mechanisms of human pancreatic cancer cell CD44 protein expression; TNF-α and IFN-γ may be involved in the process of tumor invasion and metastasis by up-regulating the expression of CD44 variant protein in tumor cells.
